I tried a simple webpart in VS2008 with .net 3.0 and it has worked. I didn't go with 3.5 as am not sure if clients have this installed and to ask them to install 3.5 will require a change request. Though as you say it's additional dlls, so if we don't use those dlls we should be fine with not having 3.5 on clients server or do we require it to be installed.
