No.

When you specify:

   X_MOUSE_DEVICE = /dev/ttyS0   (or S1)

The serial.o kernel module will be automatically loaded, which will
make the /dev/ttyS0 device node magically appear in the dev directory.

This is assuming that you've installed the ltsp_kernel package, which 
includes all of the kernel modules that you'll need.
