If I understand your question correctly, I think you’re halfway there with
your example file by using the enumitem module/package.

Use Enumerate (numbered list) rather than Itemize (bullet list) to let
LyX/LaTeX deal with automatic numbering.

Then add the enumerate options to customize the first item in the list (as
you’ve done).

(See the Users Guide > sections *3.3.7.1 Custom Enumerate Lists* and *3.3.7.3
List Spacing* for all the gory details.)

Attached is an example file (LyX 2.4.4 on Windows) which might point you in
the right direction.

Also, just for information, in your file the display equation in the second
item is in its own paragraph, which will mess up your numbering; you should
join the display equation with the text above by removing the return
between them.
