Quick answer:

goto start/programs/miktex/miktex options

a window will popup.  pick roots/add, and browse to find the LyX
directory.  browse till you select share/lyx, click OK to everything,
and you're all set...

I encourage you to skim through the FAQ files in the MikTeX
distribution, at least  (start/programs/miktex/help).  You might as
well subscribe to the MikTeX user mailing list.
