I haven't tried this, but I don't see why LyX should need to be on the same drive as MiKTeX, as long as all paths are correctly set. You might try starting LyX from a command prompt with 'lyx -dbg latex' and see if attempting to View->DVI or File->Export->DVI generates any helpful messages.
