It *must* be a problem with LyXs call of aspell. I tried everything, I can think of... I give up now! Under Linux it works very fine without any problems. But Windows? It is a pity!

Then try this:

1. unistall your manually installed Aspell, all Aspell dictionaries
   (even if we know that some things will remain)
2. delete the remaining aspell folders manually
3. check that aspell is not listed in the PATH environment variable of Windows
4. check the registry if there are still some Aspell entries although you uninstalled it, if so, delete these entries but be careful not to delete anything else!
5. uninstall LyX completely and reinstall LyX using this installer:
   http://wiki.lyx.org/Windows/LyXWinInstaller
   Open an internet connection before you start installing!

That's it. Now you should have a working Aspell installatuion with at least the dictionaries for English and the Windows system language (in your case Czech or German I think).
