I am using Lyx 1.6.5, MikTeX 2.8. But I have it installed on a machine with
no internet access, and I think that the MiKTeX manager needs internet to
find the esynt file. Where can I find this file myself? And how do I install
it?

Would be nice if you wrote the name and version of your LaTeX
distribution. Is it MikTeX? There's a package manager where you can find
esint.sty.
The other option would be to abandon using esint (I personally don't like
the changes it introduces in integral signs):
Document Settings > Math Options > Use esint package automatically
