I don't think you can hide messages you've read. But getting rid of the preview 
window; here's how I think it's done with VoiceOver.

Interact with the splitter next to the Preview window and drag the size down to 
0. Then you have to do a physical mouse click on that splitter I think. Move 
the mouse pointer to the splitter using Command+VO+F5 and then use the trackpad 
to click it. That's just from memory; but I think it works.
