Without knowing anything about smisk, I would say you need to make three different ports. If the port was called "py-smisk", then it should only be for python 2.4 by convention.
Personally I would prefer to have a way of installing one package but getting support for several python versions. Dunno if variants would be the best way though (as the package would need to be part of one of the python groups).
