With the addition of sudo, I get:

COMMAND PID USER    FD   TYPE DEVICE SIZE/OFF                  NODE       NAME
mysqld         79  _mysql 10u unix   0x40bdd8b3f53719a7        0t0            
/opt/local/var/run/mysql56/mysqld.sock

but there is still no sock file at that location. As I mentioned earlier, the 
mysql-error log does show the creation of the sock file on the last line after 
restarting.

Is there a process that might destroy it after it's created?

I should have included 'sudo' before that command because you do need to run it 
as root. Like this:

        $ sudo lsof -U -a -p $( pgrep mysqld )
        Password:

I am mystified. I installed mysql57 with MacPorts for the first time to look 
into this problem and didn't run into the missing socket problem nor did I get 
that hidden file:

        $ ls -la /opt/local/var/run/mysql57/
        total 8
        drwxr-xr-x  4 _mysql  _mysql  128 Mar 29 00:41 .
        drwxr-xr-x  5 root    wheel   160 Mar 27 15:38 ..
        srwxrwxrwx  1 _mysql  _mysql    0 Mar 28 22:48 mysqld.sock
        -rw-------  1 _mysql  _mysql    6 Mar 28 22:48 mysqld.sock.lock
