In Mail, press cmd-comma, then choose the Signatures button from the toolbar. Pick an account from the table, then a signature from the table of signatures for that account. If you've never added any, this second table is empty. To add a new signature, choose "add" after the table. To edit one, select it from the table and move to the text field holding the signature's text. Once you're done, simply press cmd-w to close the window.
