There are a couple. First, no 5S, no Alex.
Second, you have to download it. Go into settings / general / accessibility / speech / voices and select it to download. Bear in mind, it's 869MB so will take a while. Next, you'd probably have to switch your default voice to be Alex in Voiceover. Even if it says default is Alex, it will still talk with Samantha's voice.
