OK On the Macintosh open the Messages App. (Why would you think you could do
anything with iTunes for this? Messages has nothing to do with playing music.?)

Once in the messages app move over to the list of messages since it is the only
table on the aap screen, I usually use control-option-command-shift-T.
Now interact with that table.
Find the muted conversation and use control-option-shift-M to bring up the
contextual menu and you should find "Do not disturb" checked. Uncheck the item
and the conversation should now be unmuted.
Note, it might be possible to mute a person in the contacts application.
