All of Growl's stuff is in System Preferences. You can set up how it acts by default and how it works with specific applications.
Applications that can use Growl will register with it automatically. If you choose the Applications tab in Growl Preferences, you can see each application that has registered with Growl. You can blanket turn on and off all notifications for that application by checking or unchecking the item in the applications list, or select one and press Configure to fine-tune the notification settings for that app.
Note: Don't check Hide All Notifications in the general Growl stuff, otherwise you won't get any notifications. I guess that's a good way to temporarily mute it.
