I don't think you can do this natively in OS X.  You can use disk Utility to 
create an encrypted disk image of a folder.  FileVault encrypts the entire 
system disk.  You could lock a file or folder or change it's permissions to 
grant no access, but I am not aware of a way to place a password on a file or 
folder.  However, I surely could be wrong and there may be third party 
solutions for this.
