Where on the word are you? Also, are you possibly mistaking the "misspelled word" sound for the "suggestions available" sound? The former is the sort of ratchet sound, the latter a higher, shorter tone. Finally, do you have "correct spelling as I type" enabled in system preferences and in the application? I had it on in system preferences, but for the longest time it would not work in Mail.
