Activities are unfortunately named, as they do not do what they sound like. An activity is a way of associating certain settings with certain applications. For instance, you might have an activity called "edit", in which vo is set to speak text attribute changes, use phonetics, and echo every character and word typed. You can choose it manually (vo-x) or have it auto-enable itself when you enter an app of your choosing, such as Text Edit. Once you leave the app, or use vo-x to choose your default activity, those settings go away and your normal settings return.
One thing to note about activities is that they can sometimes cause the Mac to slow down when you switch apps, since VO has to load the activity from disk each time. Of course, if you have a lot of ram and/or a flash-based hard drive, you may notice little to no difference, but it is something to keep in mind. Scripts are different: you run them with commanders. For example, I have my keyboard commander set to speak time/date, ram status, wifi status, and so on, but I could as easily have assigned some of those scripts to the trackpad or numpad commanders if I wanted to. Of course, scripts can do a lot more than just speak, but your use case will differ from mine and I can't help further unless and until you have questions specifically about scripts. You can run scripts a couple other ways as well, but those are not VO-specific. Again, I don't know your use case for scripts so can't offer much more for now.
