There are 2 (orthogonal) ways to do it: either set this option for a specific folder (and then select -- wihtout necessarily opening -- it before composing a message) or create a new identity and set Reply-To value for it. In both cases the option itself is in the "compose" page of the options dialog. Personally I use both: for quite a few of my folders I have specific Reply-To set and I also have a couple of identities which I use when composing new messages.
