You could try coding it as an HTML tag, e.g. <a href="%(user_optionsurl)s">Unsubscribe</a> or whatever you do to generate the URL. Of course this will be pretty ugly in the plain text part and perhaps not recognized as a URL by an MUA that displays that part and renders recognized URLs as clickable in plain text.
