I've been having horrible problems with this as well. For a while, only using -R would focus the reply pane (otherwise I'd have to click in the reply box, wait, then click again; in safari, I'd click and start typing with no problem). With the current beta, even - R doesn't focus. I have to do the two-click thing.
It does work for me in Safari and FF, so this is a mailplane-specific problem.
