I have the same issue mentioned by Bradley, on both a Gmail account and a Google Apps for Domain account, with 2 factor authentication turned on.
It looks like the wrong password is used by Mailplane, and I have to input by hand the correct password each time I open Mailplane. Another good reason for never closing Mailplane! :-) There is obviously something broken in the way Mailplane manages the 2 factor authentication, not a big showstopper but definitely something that could be fixed.
