Colin, I have more than a few Gmail and Google Apps for Domain accounts that use 2-factor authentication, all present in my Mailplane configuration.
The only issue I have is the need to type the web password once Mailplane starts, but then I keep the second authentication cached for 30 days and have no problem to log in or switch accounts. Surely it will be better when 3.0 comes out, but for now it's not a terrible issue, in my opinion.
