Try to enter http://localhost/mapguide/mapagent/mapagent.fcgi in your browser. Where localhost is the name of your server and mapagent is the name of your application (open the IIS manager to check the name). If you get an error message, that could lead you to the problem. A common problem is that CGI is not enabled/installed in IIS or that the handler mappings are not set correctly. Try to follow this guide: http://trac.osgeo.org/mapguide/wiki/MGOS2onVistaIIS7 The latest version btw. is 2.2, you should be using that instead. The front page is a bit outdated.
