If you installed MapGuide on the cloud server with the same configuration, then yes using the cloud server IP address should work.
As to which one to operate? In software development, we have the notion of a dev/test environment and a production environment. Assuming the cloud server is your production environment, then I'd say you should do the bulk of your authoring/development on your localhost where you can experiment and play around.
