Hi Set the DEFRESOLUTION to 72 (I assume all your mapfile settings are for a 72dpi image) , change the RESOLUTION to 300. Change your map size if you want a big 300dpi image.
It is possible that the image header doesn't contain the DPI information. This can easily be modified with a graphic program like "The GIMP".
