One mapfile (.map) defines one map.
 
You can of course write as many mapfiles as you like, therefore defining many maps.

I am new to Mapserver.  I was wondering if anyone knows if it is possible to have multiple maps generated in Mapserver?  If so, Does this require multiple .MAP files?
