Hi there Jennifer, Just on the off chance, are you trying to create overviews in your tiff or are you really trying to create internal tiles? My suspicion is that you really just want overviews (aka pyramids in the esri lingo) to speed up Mapserver rendering...
If that is the case then you should looks at gdaladdo and specify the levels you want the overviews to be created.
