The only viable part of checking the trans fluid while cold is knowing that there is enough fluid in the pan to keep things reasonably lubricated. I'm sure Mercedes has a specific reason for the outlined procedure.
(off the top of my head...)
1. Drive until the transmission is fully warmed up.
2. park the car on flat ground.
3. idle in each gear (P R N D) for 5 seconds each
4. Check fluid level
5. add/remove fluid as needed
6. repeat 3-5 as needed
