you can do that, but you would also concentrate all the other stuf in your sample. The Wessel & Flügge method, and also Bensadoun & Weinstein (DOC + TCA) have the advantage that you remove some interfereing materials like salts, detergent etc. Also, they are quicker than lyophilisation and easier to handle with a large number of samples.
