I have a lentiviral system working in my lab (pInducer), my PI wants to know if we could use a pBabe based vector as well. pBabe apparently has MMLV based LTR instead of HIV. My question is whether the packaging constructs I have for lentiviruses will work for MMLV?
I'm guessing that HIV and MMLV have different capsids, which would mean that gag would be different. Since gag and pol are on the same plasmid, I have to replace both of them. What about the envelope? I know you can switch envelopes within a retroviral system. (e.g. pRetroX has VSVG, Ampho, and Eco). Can you switch *between* systems? Is the VSVG I'd use on MMLV different from the one I'd use with HIV? I'm finding a lot of stuff that talks about gag pol and env, but nothing that's really explicit about what they actually do. e.g., what part recognizes the LTR? Is that in 'pol'?
