So there is already something running on your listen port, netstat and ps might 
help.

Snapshot of OpenBSD or snapshot of OpenSMTPD? 
Which package of opensmtpd-extras? 

You need the latest snapshot tarball or latest -current opensmtpd-extras 
package for it as well.
Otherwise filters will not work, because path for loading them has changed 
recently.

If you need hints what is going wrong start with debug/verbose mode, e.g.: 
smtpd -dv
