Are you using the latest version 2.2.1? Did you follow the instructions here:

Hey guys, Been using Mnemosyne a lot now for Pharmacology and travel from one computer to another to do my studies often. I have Mnemosyne Portable on my USB drive. Every time the drive letter changes I receive this type of error. At the moment, I have to uninstall and reinstall Mnemosyne to alleviate the problem. Complicated question, is there a fix?
