Check if there's a file sound001.mp3 in
<mnemosyne_data_dir>/default.db_media (if you used the default name
for your database) and see if you can play that file outside of
Mnemosyne. If you inserted sound through the menu you get when
right-clicking, that should be fine.

Otherwise, you could try upgrading to the latest version
https://mnemosyne-proj.org/download-mnemosyne.php
