I've used TestFlight with 5.2.x and 5.3/5.4.x, and no issues with it at all. iOS6 or the MonoTouch version should have no effect on it at all.
It needs iOS4+ tho, but thats normal. Over the air updates/installs didn't exist in iOS3. Try it out - it's free and fairly easy to setup.....
