You'll do it for all specimens. Then you either load the file in TPSUtil and specify the semilandmarks as explained in the help file or you write a slider file manually. Say that semilandmarks are landmarks 10 and 11 and they have to be slid 'between the boundaries' set by 9 and 12, then your slider file will be (if I remember well how to make them!): 1 2 3 0 9 10 11 10 11 12 Save the txt file with nts extension and you're done.
