I suspect this is because the rest of the 'Target' contains a space, e.g. "C:\Program Files\mozilla.org\Mozilla\Mozilla.exe". When all that's in the target is a full file path, Windows doesn't mind the space, but when you add an option, it's gets all confused... (poor Windows :) ).

All you need to do (if this is the problem) is put double-quotes around what was in the target box before you added the -nosplash option, so, instead of

C:\Program Files\mozilla.org\Mozilla\Mozilla.exe -nosplash

you want to have

"C:\Program Files\mozilla.org\Mozilla\Mozilla.exe" -nosplash

Also, in answer to your other question of startup speed, removing the splash screen doesn't (as far as I know) affect how quickly Mozilla starts up, it just distracts you while it takes ages... ;)
