http://www.xulplanet.com/ is a good place to start if you want to write extensions to Mozilla applications. It explains about XUL, the XML application to define user interfaces, how JavaScript interacts with XUL and XPCOM components mostly written in C++ to provide the glue.

Note that I can't say for sure whether what you find there applies to Thunderbird as I have not even used Thunderbird so far but in general the Mozilla suite and the stand-alone applications like Firefox and Thundebird share XPCOM components.
