[MP3 ENCODER] Matlab Specgram function

Hi,   I am seeking advice on how to use matlab to make a spectrogram of a .wav file.  Do parameters such as stereo/mono, sampling rate matter etc? If anyone has experience in this procedure could they please advise me.    Thank you A. Jones
