Any recoding of an MP3 loses quality, so keep your MP3 as they are. If
you have WAV input (e.g. from a CD), then let LAME choose what it thinks
is best (Stereo/J-Stereo).
Different Quality settings may sound different, depending on the quality
of your listening equipment and your ears. Don't expect a different
sound on ordinary PC speakers. A different quality setting also changes
the filessize for VBR encodings.

It's not possible to answer this. This depends on your preferences. I
don't know about the possibilities of the GUI you use, I use LAME from
the command line and encode with VBR and "--alt-preset standard". Maybe
you want to visit www.hydrogenaudio.org and read a little bit about it.
