Definition Updates are only released three times a day to Windows Update/Microsoft Update (and thereby SCCM Update Points), while new definition updates are released continuously to the Malware Protection Center.
If your run MpCMdRun.exe (the built-in updater for Windows Defender/Endpoint Protection) with the switch -SignatureUpdate -MMPC you'll receive updates directly from the Malware Protection center. However I'll assume the delay in releasing updates thru the common channels is to allow a timeslot for Microsoft to do QA/Regression testing on the new updates, so it would be unwise to roll out updates directly from the Malware Protection Center.
