there are so many versions and locking options of both mutt and gnus
that nobody will be able to answer that questions without a very thorough
look at your OS and configuration.
In practice both try to do correct locking so you could be lucky.

More serious question - do they even use the same mbox format? Some time 
in the past gnus used some special format. Mutt is famous for beeing able
to use pretty much every format but I would double check this.
