No, usually you don't need to. AFAIK the API is enabled by default and should work out of the box. Maybe you configured something (in MediaWiki or in the web server setup) that breaks it? As I already noted: your api.php does work for some queries, so it's in enabled and accessible in principle, but it doesn't return the wikitext of an article. I noticed that you're using some fancy URL-rewriting, maybe that has something to do with it? That's really only guessing though...
