I think you can do a whole subnet at one time using your network address. 192.168.1.1/24 Or whatever yours happens to be. However, I would suggest doing ranges of IP's rather than the whole subnet. You can drastically slow the network and even cause some deivces to reset themselves or force you to reboot them. There is an option for Safe Check that will make the scan a little safer, but you still eat a lot of bandwidth and really put a load on any switches you may have and cause them to reset.
If you are doing this at work, I would wait until the weekend when nobody is there if you want to do the whole subnet at one time. Even if yoiu do this with a range of addresses, depending on the scan options you choose and the range of addresses, you can still hurt the network. If you want to do a range of address, you can enter them as 10.2.12.1-25 Or you can create text files with ranges of address and load them into nessus. The systems that you scan depend on what plugins you use. If yours are not up to date you can run "nessus-update-plugins -v" from the command line. But again, I would do it a little at a time.
