Knoppix images are easily updated, you just need a Debian system with enough space (~8GB of free space to create the images for the full Knoppix version and less for the -lite one). You could take the Nessus packages I uploaded to http://people.debian.org/~jfs/nessus and install them in a chrooted environment instead of the ones provided (1.2.7 IIRC), then remake the ISO image. This should updated both the plugins and the engine.
I can point you to HOWTOs/references on the exact procedure if you are interested.

PS: I find Knoppix really cool and I would like to have a Knoppix-based CD providing updated security tools (such as Nessus, which already is) and more already provided by Debian (http://www.debian.org/doc/manuals/securing-debian-howto/ch-sec-tools.en.html)
in the same way the FIRE Cd (former Biatchux) is provided.
