Hi Matt,
Latest NfSen snapshot on sourceforge supports adding/deleting new netflow 
sources to profile live.
If you do not want to upgrade for any reason have a look into the file 
etc/nfsen-shell-param.
There is a line SOURCES, which list each source:port which is needed for nfcapd 
startup. This should do the job.
