Even a tiny amount "open" will cause a very poor idle.

Up should be choked, I believe it's actually a valve to deliver more fuel 
rather than restricting air like a traditional choke.

I should have said "pilot screw". It's the one with the limiter cap on it 
normally and will be centered side to side under the forward flange of each 
carb 
on the CB650SC. (Some of us old folks refer to the pilot screw as the idle 
mixture adjustment screw)
