If you already have significant background writing modules then I'd start in the issue tracker. Look at both open and closed issues to see how we usually go about fixing stuff. Also use it to guide how you learn the internals. It's helpful to have a specific goal in mind.
If you don't understand a specific area then start by writing a module with the API.
