One can only get spot colours for a bitmap into a .eps (or .pdf) or .psd.
What one could do using libtiff is split the channels of the file, toss magenta and cyan, then use a different tool to composite the two bitmaps, assigning the yellow bitmap to the spot colour --- FreeHand could do this via scripting, if ConTeXt can place a bitmap on a particular spot colour plate and control overprinting of bitmaps, one could handle the balance in it.
