I believe that the Internet Connection is not REQUIRED on the target machine, but if it has one then System Sweeper will grab updates. Without an Internet connection System Sweeper will just scan/clean with the definitions it has on-hand from when it was burned.
It's probably a good idea to burn new System Sweeper discs every couple of months if you are frequently cleaning machines that don't have Internet connections otherwise you'll just have a progressively older version of Sweeper.
