They will expire 60 days from when they were last changed. That means that if all your user's passwords were last set a year ago, and you set this policy today, they will all be forced to change next time they logon.
The mitigation plan for this is to first mark password never expires on every user and then release the accounts for reset in batches (by unchecking the box/clearing the bit).
